Project Workflow
Complete overview of the Adaptyv Bio project workflow from experiment configuration to final reporting
Experiment Configuration
The process begins when you submit your project details through the Foundry Portal. Our intuitive configurator allows you to specify:
- Type of experiment: Choose from binding screening, affinity characterization, epitope binning, and more
- Protein variants: Upload sequences or select from our library
- Target molecules: Define the antigens or ligands of interest
- Custom requirements: Set specific concentrations, controls, or replicates
After submission, our team reviews the feasibility of your project, creates a detailed project plan, and communicates any necessary adjustments or recommendations.
Build Phase
During the build phase, we prepare all necessary materials and reagents for your experiments. This includes:
DNA Synthesis and Optimization
- Synthesis of DNA sequences encoding your protein variants
- Codon optimization for efficient expression
- Quality verification of synthesized constructs
Protein Expression
- Production of protein variants using our cell-free expression system
- High-quality, consistent protein yields
- Verification of expression levels and quality
Target Preparation
- Sourcing or production of target molecules
- Access to our pre-curated antigen library or custom target preparation
Buffer and Reagent Preparation
- Tailored to your experimental conditions
- Optimized for stability and performance
Data Generation (Protein Testing)
This is where we test your proteins and targets using the experimental workflows you’ve selected. For binding experiments, we use bio-layer interferometry (BLI), a label-free technology that provides real-time binding kinetics.
Experiment Execution
- Immobilization or presentation of proteins on biosensor tips
- Introduction of target analytes in solution
- Real-time monitoring of binding interactions
Technical Precision
- Single or multiple technical replicates based on your requirements
- Consistent experimental conditions across all samples
Controls
- Inclusion of negative controls to assess non-specific binding
- Positive controls to validate experimental setup
Quality Assurance
- Continuous monitoring of experimental parameters
- Flagging of unexpected results for review
Analysis, Validation, And Reporting
After data acquisition, we perform rigorous analysis and validation to ensure the accuracy and reliability of your results.
Data Processing
- Fitting of raw sensorgrams to appropriate kinetic models
- Extraction of key parameters (k_on, k_off, K_D values)
- Statistical analysis of replicate data
Validation
- Cross-checking results against controls
- Verification of data consistency across replicates
- Identification of any outliers or anomalies
Comprehensive Reporting
- Detailed report including:
- Raw data and processed results
- Kinetic parameters and binding affinities
- Experimental conditions and notes
- Interpretation and recommendations
Foundry Portal Access
- View and download raw sensorgrams
- Explore fitted kinetic models
- Filter and categorize results

Frequently Asked Questions
What is the typical end-to-end workflow for an experiment?
What is the typical end-to-end workflow for an experiment?
Our workflow is designed to be simple and transparent:
- Setup & Submission: You create an experiment in our Foundry Portal, select your target, and upload your binder sequences for review.
- Confirmation & Quote: We confirm the experimental details and provide a quote for your approval.
- Execution: Upon your approval, we handle everything: DNA construct design, gene synthesis, protein expression, and running the assays.
- Data Delivery: We process the data, perform quality control, and upload the final results to the portal for you to access and download.
We will notify you by email as your project progresses through these key stages.
What are the requirements for submitting protein sequences?
What are the requirements for submitting protein sequences?
Submitting sequences is straightforward. You only need to provide the amino acid sequence for each of your binders; we handle the rest, including DNA design and codon optimization for our E. coli cell-free expression system.
Key requirements:
- Initiator Methionine: You do not need to add an initiator methionine to your sequences; our system will add one if it’s missing.
- File Formats: The Foundry Portal directly accepts uploads in CSV and FASTA formats. If your sequences are in another format like an Excel spreadsheet, you can send the file to our team via the in-app chat, and we will upload it for you.
